RNA travels with ZBP1

چکیده
pposites really do attract. Results on page 101 by Oliveria et al. show that, although they perform opposing functions, a kinase and a phosphatase linked to synaptic plasticity are close neighbors at the plasma membrane. The two are brought together by the scaffolding protein AKAP79 into a complex that may be important for efficient learning. The close proximity of the three proteins is uncovered by a powerful microscopy technique, fluorescence resonance energy transfer (FRET). Using this technique in COS cells, the group shows that protein kinase A (PKA) and the phosphatase calcineurin (CaN) bind to sites on membrane-targeted AKAP79 that are spaced only nanometers apart. In neurons, AKAP79 is known to O

Sensing of viral and endogenous RNA by ZBP1/DAI induces necroptosis
Nucleic acids are potent triggers for innate immunity. Double-stranded DNA and RNA adopt different helical conformations, including the unusual Z-conformation. Z-DNA/RNA is recognised by Z-binding domains (ZBDs), which are present in proteins implicated in antiviral immunity. These include ZBP1 (also known as DAI or DLM-1), which induces necroptosis, an inflammatory form of cell death. Using re...
متن کاملSpatial regulation of beta-actin translation by Src-dependent phosphorylation of ZBP1.
Localization of beta-actin messenger RNA to sites of active actin polymerization modulates cell migration during embryogenesis, differentiation and possibly carcinogenesis. This localization requires the oncofetal protein ZBP1 (Zipcode binding protein 1), which binds to a conserved 54-nucleotide element in the 3'-untranslated region of the beta-actin mRNA known as the 'zipcode'. ZBP1 promotes t...
متن کاملLimited availability of ZBP1 restricts axonal mRNA localization and nerve regeneration capacity.
Subcellular localization of mRNAs is regulated by RNA-protein interactions. Here, we show that introduction of a reporter mRNA with the 3'UTR of β-actin mRNA competes with endogenous mRNAs for binding to ZBP1 in adult sensory neurons. ZBP1 is needed for axonal localization of β-actin mRNA, and introducing GFP with the 3'UTR of β-actin mRNA depletes axons of endogenous β-actin and GAP-43 mRNAs a...
متن کاملFeedback regulation between zipcode binding protein 1 and beta-catenin mRNAs in breast cancer cells.
ZBP1 (zipcode binding protein 1) is an RNA-binding protein involved in many posttranscriptional processes, such as RNA localization, RNA stability, and translational control. ZBP1 is abundantly expressed in embryonic development, but its expression is silenced in most adult tissues. Reactivation of the ZBP1 gene has been reported in various human tumors. In this study, we identified a detailed ...
متن کاملZBP1 recognition of beta-actin zipcode induces RNA looping.
ZBP1 (zipcode-binding protein 1) was originally discovered as a trans-acting factor for the "zipcode" in the 3' untranslated region (UTR) of the beta-actin mRNA that is important for its localization and translational regulation. Subsequently, ZBP1 has been found to be a multifunctional regulator of RNA metabolism that controls aspects of localization, stability, and translation for many mRNAs....
متن کاملSpatial arrangement of an RNA zipcode identifies mRNAs under post-transcriptional control.
How RNA-binding proteins recognize specific sets of target mRNAs remains poorly understood because current approaches depend primarily on sequence information. In this study, we demonstrate that specific recognition of messenger RNAs (mRNAs) by RNA-binding proteins requires the correct spatial positioning of these sequences. We characterized both the cis-acting sequence elements and the spatial...
